چکیده

The kinetics of rhodopsin photolysis in sonicated suspensions of rod outer segments (R O S), unsonicated ROS suspensions and rhodopsin detergent solutions were compared. No significant kinetic differences were detected; however, the energy of activation for the metarhodopsin I to metarhodopsin II reaction was somewhat higher in sonicated ROS suspensions than in the un sonicated suspensions. W hile the sonicated preparations contained glycerol, it was not possible to show a chemical influence of glycerol on the reactions of the rhodopsin decay. An often assumed effect that the ROS outer membrane acts as a barrier toward diffusion of protons could not be confirmed.

Rapid calcium release and proton uptake at the disk membrane of isolated cattle rod outer segments. 1. Stoichiometry of light-stimulated calcium release and proton uptake.

We reported a rapid, light-stimulated release of calcium from isolated rod outer segments that is apparent only when both the disk membrane and the plasma membrane are made permeable to calcium by adding the ionophore A23187 [Kaupp, U. B., Schnetkamp, P. P. M., & Junge, W. (1979) Biochim. Biophys. Acta 552, 390-403]. In this paper, we have investigated the light-sensitive diskal binding sites a...

Temperature and pH dependence of the metarhodopsin I-metarhodopsin II equilibrium and the binding of metarhodopsin II to G protein in rod disk membranes

The equilibria between metarhodopsins I and II (MI and MII) and the binding of MII to retinal G protein (G) were investigated, using the dual wavelength absorbance response of rod disk membrane (RDM) suspensions to a series of small bleaches, together with a nonlinear least-squares fitting procedure that decouples the two reactions. This method has been subjected to a variety of theoretical and...

Structure and function in rhodopsin: the fate of opsin formed upon the decay of light-activated metarhodopsin II in vitro.

We report that the light-activated bovine metarhodopsin II, upon decay, first forms opsin in the correctly folded form. The latter binds 11-cis-retinal and regenerates the native rhodopsin chromophore. However, when the opsin formed upon metarhodopsin II decay is kept in 0.1% dodecyl maltoside, it converts in a time-dependent manner to a form(s) that does not bind 11-cis-retinal. On subsequent ...

Rhodopsin photoproducts and rod sensitivity in the skate retina

The late photoproducts that result from the isomerization of rhodopsin have been identified in the isolated all-rod retina of the skate by means of rapid spectrophotometry. The sequence in which these intermediates form and decay could be described by a scheme that incorporates two pathways for the degradation of metarhodopsin II (MII) to retinol: one via metarhodopsin III (MIII) and the other ...

Function of the farnesyl moiety in visual signalling.

The aim of this work was to search for the biological function of protein isoprenylation. For this purpose, peptides were synthesized and, by using a convenient protocol, were farnesylated or geranylated at the thiol group of the C-terminal cysteine. The interaction of these peptides with photoactivated rhodopsin (Rho*, which is functionally equivalent to metarhodopsin II) was studied with the ...
